Maybe broaden the muzzle slightly at the nose end. The muzzle seems to terminate with just a nose too much like a cone. I think the overall look is good and I think the eyes are plenty big enough. It's a matter of personal taste.

I think that whatever you use, try to stick with it as you will get super confortable with it and you don't want to have to relearn with new programs over and over.

I haven't done one either, but it would depend on what your comic is about. Is it going to be a series of single stand-alone jokes or a strip with continuity? If you do a strip, then I highly suggest you draft your main story line arc and rough in your characters, etc. Nothing worse than...
